The major goal of this research project is to understand in-depth mechanistic and structural aspects controlling catalysis by p-hydroxyphenylacetate hydroxylase and pyranose oxidase. We will use techniques and methodologies that are specialties of our group and well established for usage, i.e., stopped-flow spectrophotometry, rapid-quench techniques, absorbance/fluorescence spectroscopy, kinetics and thermodynamics, as well as a variety of enzymological tools. In addition, collaborations with leading scientists in the field (see project links and collaborations) will enable us to use X-ray crystallography, molecular dynamics simulations, and possibly single-molecule spectroscopy to study the questions being addressed.
