This project is aimed to develop highly active regulatory sequences in cyanobacterium Synechococcus PCC7942 by generating strong promoters and optimal non-SD initiation of translation sequence from synthetic DNA libraries.
1. To construct the synthetic promoter library.
2. To screen and analyze the synthetic promoter activities in Synechococcus.
3. To analyze the relationship between the synthetic promoter sequences and the
promoter activities in Synechococcus and E. coli.
4. To construct the synthetic non-SD sequence library.
5. To screen and analyze the synthetic non-SD sequence activities in Synechococcus.
6. To analyze the relationship between the synthetic non-SD sequences and the reporter activities in Synechococcus and E. coli.
