วัตถุประสงค์
:
1 First specific aim is to examine the effect of tetrahydrobiopterin inhibition on NO-mediated response of coronary arteriole. The NO-mediated vasodilation was examined in isolated and pressurized (60 cmH2O) porcine coronary arterioles (50-100 ?m) before/after treatment of BH4 inhibitor (DAHP, 10 mM).
2 The specific involvement of various reactive oxygen species during uncoupled eNOS will be determined using physiological and cytochemical approaches.
3 To investigate the compensatory mechanisms for the NO-mediated vasodilation during the eNOS uncoupling.
