The objectives of this study are to select P. monodon families that could survive and grow well when being reared in low salinity water, and to find the mechanism that could explain that phenomenon. Specifically, the proposal includes a comparison study in the gills and antennal gland of P. monodon reared under 10 and 20 ppt seawater on the following items:
1. Ultrastructure by transmission electron microscope (TEM)
2. Localization of Na+/K+-ATPase by immunohistochemistry
3. Determination of Na+/K+-ATPase activities by measure inorganic phosphate liberated
4. Determination of Na+/K+-ATPase protein by SDS-PAGE and immunoblotting techniques
5. Determination of the mRNA expression of Na+/K+-ATPase gene by semiquantitative reverse transcriptase polymerase chain reaction (RT-PCR) technique
