วัตถุประสงค์
:
1. To clone and over-express the gdh genes from Bacillus sp. SB5 and B. subtilis 168 in the host-of-interest, an organic solvent-tolerant B. subtilis 3C5N.
2. To study the kinetics properties of glucose dehydrogenase (GDH) from an organic solvent-tolerant Bacillus sp. SB5 and compare with that of B. subtilis 168.
3. To evaluate the potential applications of GDH from Bacillus sp. SB5 as an isolated enzyme for an in vitro coupling reaction (coupling with an enzyme P450 BM3 F87V) as well as a permeabilized whole-cell biocatalyst.
