วัตถุประสงค์
:
1. To examine the expression of Aurora kinase family members including Aurora-A, Aurora-B and Aurora-C, as well as p73 isoforms in p53-mutated oral squamous cell carcinoma cell lines, ORL-48T and ORL-136T as compared to the normal human gingival epithelial cells (HGEPp.05, CELLNTEC, Bern, Switzerland) at the mRNA and protein levels.
2. To investigate the TAp73-mediated induction of apoptosis by inhibition of Aurora kinases with specific Aurora-A kinase inhibitor, specific Aurora-B kinase inhibitor and pan-Aurora kinase inhibitor, in p53-mutated oral squamous cell carcinoma cell lines, ORL-48T and ORL-136T. The protein expression of pro-apoptotic TAp73 and its downstream targets, including Bax, and p21WAF1, will also be investigated.
3. To investigate whether TAp73 is important for Aurora kinase inhibitor-induced apoptotic cell death using siRNA against TAp73
