วัตถุประสงค์
:
The ultimate goal of this work is to study the therapeutic potential of splice-switching oligonucleotides, specifically peptide-conjugated phosphorodiamidate morpholino oligomers (PPMOs), for ?-thalassemia in the mouse model. The specific aims are:
2.1 To evaluate the splicing correction efficiency of candidate PPMOs in erythroid mononuclear cells isolated from bone marrow of the IVS2-654 ?-thalassemic mice. The properties of erythroid cells after treatment will be assessed compared to untreated cells and cells isolated from wild-type mice.
2.2 To evaluate the splicing correction efficiency of candidate PPMOs in vivo in IVS2-654 ?-thalassemic mice. The hematological parameters will be assessed after treatment.
