วัตถุประสงค์
:
2.1 To generate Nkx2.5-eGFP mouse ES cell lines.
2.2 To differentiate Nkx2.5-eGFP mouse ES cell lines into Nkx2.5-eGFP CPCs and cardiomyocytes.
2.3 To further purify and enrich Nkx2.5-eGFP CPCs derived from Nkx2.5-eGFP ES vs. wild type ES cells by using FACS and subsequent culture them into relevant functional mature cardiomyocytes.
2.4 To investigate the effects of cardioactive drugs on Nkx2.5-eGFP CPCs and mature cardiomyocytes derived from Nkx2.5-eGFP mouse ES cell lines, an in vitro model for studying cardiac physiology and pathophysiology.
