วัตถุประสงค์
:
Utimate Objective
To construct CAR-T cells that target the HCV protein(s) on the surface of HCV infected cells that are cytotoxic against HCV infected cells and enhance antiviral cytokine production.
Specific objectives
1. To produce and purify recombinant HCV protein(s) and produce polyclonal antibody against the protein(s)
2. To produce HCV from HCV cell culture system (HCVcc)
3. To detect HCV protein(s) expression on the surface of the HCV infected cells during early and prolonged infection for using as target antigen(s) for CAR-T cells
4. To select human antibody fragment (human ScFv/VH/VHH) specific to the target antigen from human ScFv or VH/VHH phage display library
5. To test binding specificity of the antibody fragment to the HCV infected cells
6. To construct CAR which target the HCV antigen from the selected antibody fragment
7. To transfect CAR to Jurkat cell line and detect cell surface expression of the CAR
8. To test the CAR transfected Jurkat cell line for their ability to produce cytokines
9. To transfect the CAR into healthy donor T cells
10. To test cytotoxicity and antiviral cytokine productive functions of the CAR-T cells against HCV infected cells
