1. To understand the nature of the dramatic secondary structural changes that take place during the key transition linked to toxicity of the binary toxin: from water-soluble to membrane-associated forms.
2. To understand the conformational changes that take place upon interaction with lipid bilayers of membrane-associated domains obtained from proteolysis and independent domain expression
3. To understand what are the determinants for binary toxin toxicity (channel formation and internalization) by performing site-directed mutagenesis, in vivo toxicity assays with mosquito larvae and in vitro assays by monitoring of membrane permeability behaviour.
