วัตถุประสงค์
:
1 To construct deletion and double point mutants in the DENV C protein and map for regions of C required for infectious dengue virus production using a virus-cell based trans-genome packaging system.
2 To analyze for the ability of wild-type and mutant C proteins to perform three interactions suggested to be important for nucleocapsid formation: C homo-dimerization, membrane association and RNA binding.
3 To design a peptide inhibitor based on the C region most needed for genome packaging, followed by testing for its ability to inhibition DENV multiplication in mammalian and insect cells. In addition, the peptide inhibitor will be tested for its interference of C interactions, i.e., homo-dimerization, host cell membrane interaction and RNA binding.
