วัตถุประสงค์
:
1) To isolate fur mutant and to construct iscR mutant in Xanthomonas campestris pv. campestris. To identify the role of Fur and IscR in iron homeostasis and oxidative stress, a fur mutant will be isolated and an iscR mutant will be constructed. These mutants will then be compared with the parental strain.
2) To characterize the expression of iscR gene or operon and fur gene under oxidative stress and in the presence of iron. It has been shown that IscR is a transcriptional repressor for its own operon (iscRSUA) in E.coli. X. campestris pv. campestris does not have Isc operon. However, iscR is located upstream of suf genes cluster (sufBCDS). This leads to questions whether the transcription of iscR cotranscribes with suf genes cluster and whether expression of iscR and sufBCDS are responsive to oxidative stress. Northern blot analysis and/or RT-PCR will be used to address these problems.
3) To study the role of Fur on iscR gene expression. It has been shown that iscR expression is regulated by Fur. In this study we will use molecular and biochemical techniques to investigate if Fur is a regulator of iscR gene/operon.
