วัตถุประสงค์
:
Specific aim 1: To apply the affinity purification method developed in the mammalian system to isolate the dengue replication machinery from infected mosquito cells
1.1. To purify dengue replication vesicle from infected C6/36 cells
1.2. To purify dengue replication complex from infected C6/36 cells
Specific aim 2: To characterize the protein and RNA composition of the isolated replication machinery from infected mosquito cells.
2.1 To identify the proteins that constitute the replication vesicles isolated from infected C6/36 cells
2.2 To identify the RNAs that constitute the replication vesicles isolated from infected C6/36 cells
2.3 To identify the proteins that constitute the replication complex isolated from infected C6/36 cells
2.4 To identify the RNAs that constitute the replication complex isolated from infected C6/36 cells
