วัตถุประสงค์
:
This project aims to culture 3D skin using biopolymer scaffolds for toxicology testing/screening of national products rather than relying on the imported skin models. This model would offer results that significantly closer to the response of native skin than a simple culture cells, and would reduce the use of animal laboratory.
The blended biopolymer scaffolds will be applied in cell culture to support skin cells forming 3D structure. The proportion of the biopolymers will be optimized to find out the best structure and properties that support cell proliferation, migration and finally from layers mimicking epidermis and dermis. On the cell culture part, keratinocytes and fibroblasts will be co-cultured on the biopolymer scaffolds. Culture conditions will also be optimized until the skin cells form 3D structure similar to native skin and be able to response to test materials (i.e. cytokine release) in the level that are comparable to commercial skin model and native skin. The final session of this project will be verification of this model with the commercial products.
