วัตถุประสงค์
:
Specific Aim#1: Identify MUC1 targeting peptide. A group of MUC1 recognister peptides will be synthesized and validated specific binding on MUC1 expressing and normal breast cancer cell lines as well as on breast cancer and normal breast tissues compared to anti- MUC1 antibody.
Specific Aim#2: Formulate and characterize MUC1-anti-cancer drug nanoparticles. MUC1 targeting peptide will be conjugated to micelle. Optimal peptide density, size, stability, entrapment efficiency (EE) of doxorubicin, release profile, and cell surface binding efficiency will be determined.
Specific Aim#3: Evaluate therapeutic efficacy of MUC1-anti-cancer drug nanoparticles as an anti-cancer agent. In vitro cytotoxicity and apoptosis assays will be performed to evaluate cytotoxic and targeted therapeutic potential of MUC1 doxorubicin micelles compared to untargeted doxorubicin micelles.
