In this study, we want to investigate the functional properties of G6PD variants found in Thai population, namely G6PD Bangkok, G6PD Songklanagarind, G6PD Bangkok Noi and G6PD Viangchan-Union. Additionally, we want to assess the hemolytic risk of G6PD deficiencies. The ultimate goal is to grant the valuable information regarding the correlation between G6PD genotype, enzyme activity and severity of hemolysis which is crucial for safe and widely use of antimalarials in individuals with G6PD deficiencies.
Specific objectives:
To construct, express and purify of G6PD variants found in Thailand
To measure the steady kinetic parameters of G6PD variants
To determine the stability of G6PD variants in different temperatures and various pH
To determine the level of reduced glutathione in coupled reaction with G6PD variants
