วัตถุประสงค์
:
To investigate the role of glutathionylation modification on the DENV NS5 RNA capping activity as it plays a crucial role in efficient viral protein replication. Therefore the project will be:
1. To construct two recombinant DENV NS5s for just the capping domain and the full-length gene, then perform protein expression and purification.
2. To identify the cysteine residues that are glutathionylated on DENV NS5 capping domain.
3. To construct cysteine/serine mutant clones, then perform protein expression and purification.
4. To study the effect of glutathionylation modification on the guanylyltransferase activity of the wild-type and mutant enzymes.
